So this video shows how to inoculate a broth from a slant So I’m starting with a slant with organism already growing on the slant, and if you can see here You can see a whitish growth that’s occurring on the surface here So I want to look for that so this tube already has organism growing on the tube So I’m going to have my sterilized tool, and I’m going to be using a loop here. I want to put that aside I’m going to remove the lid from my culture, and I’m going to put it Top down on a kimwipe a clean kimwipe hold on to that now I’m going to go to my New medium, there’s nothing growing on here. This is uninoculated And I’m going to remove that lid as well and put that on my Kimwipe. Remember which lid is in which location. So now I’m going to go and pick up my tool, which has been sterilized and cooled, and I’m just going to touch down Into the slant make sure you touch do not dig into the agar just touch down remember you’re picking up about the size of a freckle Take that out, and I’m going to transfer that over to my broth Give it a little tap you. You don’t need to shake it just a little tap I’m going to now continue to hold on to that tool and I need to put those lids back on So I’m going to pick up the lid from my starting culture Put that on and then I’m going to pick up the lid from my newly inoculated medium and then I’ll place those into my rack, and now I will go and sterilize this tool. And that is how you inoculate a liquid broth from a slant
